This novel is a parody of the Mervyn Peake Gormenghast trilogy, specifically that relating to the first two books. The elements of parody are set in terms of style and characterization rather than the specifics of the plot, although in broad terms we follow the adventures of Steelpike through the labyrinthine passageways of the castle. Many of the characters from Peake's books appear, though changed somewhat in name, sentiment and disposition. The plot departs entirely from Peake's trilogy in terms of the parallel narrative set on a different planet, this relating to the research conducted by the Doctor and the 76th Earl, Sepalcrave, this being later developed by the polymath Sophie Du Châtelet, the only significant castle based character not in the original trilogy. The cast of characters include the eponymous 77th Earl, Totus Gripe, Sourcrutch, Swetter, Spray, Rottensod, Bellend, the schoolmasters (Spratt, Welch, Snibble, Dungworth, and Deadwood), the psychotic Lady Fugue, Lady Ermentrude's sisters, Flora and Alice, and the Earl's sister, Lady Ophelia.

By 2045 the Micram Corporation's control is complete. Surveillance is near total and adherence to progressive ideology enforced with an ID chip. Apart from the use of artificial intelligence, special units of highly advanced Microids (hybrid machine/tissue robots) are deployed amongst the population. Growing evidence supports the hypothesis that many individual diseases--such as cancer, Alzheimer's, and diabetes--are actually collections of diverse syndromes that manifest through distinct biological pathways. Precision medicine is a new paradigm of care where prevention and treatment of disease is based on a common biological basis rather than on a collection of shared symptoms. Recent advances in genome editing and human pluripotent stem cell (hPSC) technology have enabled construction of clinically-relevant, genotype-specific "disease-in-a-dish" models that hold promise for furthering the development of genotype-specific precision therapies. However, these technologies have been limited by (1) the challenge of delivering genome editing components to human cells and (2) the ability to isolate precisely edited hPSCs required for development of genotype-specific disease models. In this thesis, I first describe the development and application of an image based method for optimizing non-viral delivery of CRISPR/Cas9 genome editing machinery to gene formulations. Second, I describe a method for isolating scarless, precision-edited hPSCs using transient puromycin selection. Finally, I apply these methods to study a class of inherited blinding diseases, known as bestrophinopathies, and to evaluate a CRISPR/Cas9 based gene therapy for these conditions. These methods, and their application to study of genotype-specific treatments for bestrophinopathy, will play a role in furthering the development of precision medicine for blindness and other genetic diseases.

Background In spite of lacking evidence for advantages of substitutes over titanium, it seems that there is a basic movement of patients and dentists toward the use of metal free solutions also in implant dentistry. This tendency is strongly supported by of a growing cohort of patients with concerns over titanium sensitivity, allergies or intolerances irrespective of only weak evidence for clinical relevance as well as clinical concerns regarding aesthetic outcomes in the anterior region.Aim/Hypothesis The aim of this study was to evaluate the outcomes of all-ceramic crowns on two-piece zirconia implants in a prospective randomized controlled study after up to 80 monthsMaterials and Methods 21 patients with a total of 28 implants (14 zirconia and 14 titanium) were reviewed up to 80 months following restoration. Bleeding on probing (BOP), plaque index (PI), pink aesthetic score (PES), mean radiographic marginal bone resorption, and implant survival was determined.Results After a mean observation time of 80.9 months (SD u00b1 5.5) the following data were assessed: Mean marginal bone level of -1.38mm at zirconia (SD u00b1 0.81) and -1.17mm at titanium implants (SD u00b1 0.73). BOP of 16.43% (SD u00b1 6.16) at zirconia and 12.60% (SD u00b1 7.66) at titanium implants. PI of 11.07% (SD u00b1 8.11) at zirconia and 15.20 % (SD u00b1 15.58) at titanium implants. PES of 11.38 (SD u00b1 0.92) at zirconia and 11.56 (SD u00b1 1.01) at titanium implants respectively. Two zirconia implants and 1 titanium were lost within the evaluated period after implant surgery resulting in a survival rate of 85.7% for zirconia and 93.3% for titanium implants.Conclusions and Clinical Implications In the present pilot trial no significant differences between adhesively luted zirconia abutments and all-ceramic restorations on two-piece zirconia implants could be observed after up to 80 months compared to a titanium control group. The small study population needs to be taken in consideration when interpreting the results.
