A fundamental mission of developmental biology is to understand the molecular mechanisms underlying germline determination and somatic development during early embryogenesis. This dissertation explores the function of PIWI as a maternal component in germline fate determination, mitosis, and chromatin organization during early embryogenesis in Drosophila. With regards to germline development, maternal PIWI is initially a component of the polar granule, a germ plasm-specific organelle in early embryos essential for germline specification, and then becomes localized to the cytoplasm of newly formed pole cells at embryonic mitotic cycle 10 and to their nucleus at cycle 11. Depleting maternal PIWI leads to failure in pole plasm maintenance and primordial germ cell (PGC) formation but does not affect abdominal patterning, whereas doubling and tripling the maternal piwi dose increases the number of PGCs by two- and three-fold, respectively. Furthermore, PIWI interacts with VASA and OSKAR, two key polar granule proteins, and appears to achieve its dose-sensitive control of the PGC number by regulating the levels of VASA and OSKAR. This regulation appears to be mediated by the miRNA pathway, since PIWI complexes with dFMRP and DICER-1, but not with DICER-2. Moreover, depleting DICER-1, but not DICER-2, also leads to a severe pole plasm defect and a drastically reduced PGC number. Because DICER-1 is required for the miRNA pathway whereas DICER-2 is required for the siRNA pathway, these data implicate the involvement of PIWI in the mJRNA pathway as well as in regulating the levels of OSKAR and VASA to achieve germline determination. Furthermore, maternal PIWI is essential for the development of the soma. In syncytial embryos, most of PIWI is localized to the cytoplasm during mitotic cycles 1-13 and to the nucleus after cycle 14. Embryos depleted of maternal PIWI display various mitotic defects, including cell cycle arrest, abnormal chromosomal and nuclear morphology, asynchronous nuclear division, and aberrant nuclear migration. Additionally, maternal PIWI-depleted embryos exhibit deficiencies in chromatin organization. These observations suggest that matemal PIWI plays a critical role in cell cycle progression and the maintenance of chromatin structure, with compromised chromatin integrity being a possible cause of the observed cell cycle defects.
