"Commercial and recreational fishing are important to the economy and culture of Hawaii. The deep-slope bottomfish commercial fishery preferentially targets seven high value ''Deep-7'' species (i.e, six snappers and one grouper, hereafter referred to as bottomfish). The NOAA Pacific Islands Fisheries Science Center (PIFSC) Stock Assessment Program is responsible for regularly conducting assessments of bottomfish. The stock assessment process requires reliable time-series of catches, fishing effort and life history demographics to estimate stock abundance trends and evaluate sustainability benchmarks. Current bottomfish stock assessments that rely solely on fishery-dependent abundance indices may be biased (Ault et al., 2014). A series of workshops convened in 2001-2005 evaluated existing bottomfish stock assessment methodologies in the Pacific Islands Region (PIR; Mace et al., 2001; Ralston et al., 2004). Mace et al. (2001) noted that the greatest impediment to credible bottomfish stock assessments was the lack of accurate and precise input data. To improve abundance estimates, Ralston et al. (2004) recommended development of a fishery-independent survey using available ''advanced technologies''. A 2005 workshop on Ecosystem Science and Management Planning reiterated the pressing need for a design and implementation of population-level fishery-independent survey to obtain size-structured abundance data for bottomfish, and to aggressively pursue development of length-based assessment models. This goal of this technical report was to improve stock assessments through optimal design and implementation of a bottomfish fishery-independent survey for the Deep-7 bottomfish complex in the main Hawaiian Islands (MHI) with three primary objectives: 1. Evaluate the most effective survey gears for obtaining species-specific spatial size structured abundance metrics of the Hawaiian bottomfish stock; 2. Conduct quantitative gears intercalibration studies to determine the relative fishing power of the multiple gears to be used in the survey; 3. Detail the required methodologies for efficient conduct of a multi-gear fishery-independent Survey of the MHI bottomfish stocks"--Executive summary. "Using an autonomous underwater vehicle (AUV) and towed camera system (TCS), deep-sea corals, sponges, and seafloor habitats were visually surveyed for the first time in areas of longtime trawl fishing off northern California. During an 11-day cruise aboard the R/V Point Sur 8-18 September 2014, researchers completed 6 dives with the AUV and 9 deployments of the TCS, and spent over 42 hours underwater at depths of 586-1169 meters from the Oregon- California border to the Mendocino Ridge (...) This report provides a summary of the methods and results from these underwater surveys of DSC&S off northern California. Abstract: Tumor associated macrophages (TAMs) perform various task that are essential for tumor growth, angiogenesis, metastasis, and tumor invasion. Growth factors, chemokines, cytokines and the extra cellular matrix in the tumor microenvironments signal the TAMs to perform these tasks. The tumor microenvironment is not homogeneous and can be classified into several distinct regions such as sites of tumor invasion, perivascular, hypoxic and stromal regions. Each of these sites likely has different cytokines and signaling molecules in their microenvironment that might signal and direct the function of TAMs. The purpose of this project is to isolate and analyze TAMs from distinct tumor regions to gain a better understanding of the mechanism behind the specialized role of these TAMs. As a first step towards this goal, micro array analysis was conducted on flow sorted TAMs and developmental macrophages expressing yellow fluorescence protein (YFP). Micro array data from age matched developmental macrophages was compared to TAMS from early, premalignant stage of breast cancer, and late stage metastatic breast cancer to find the genes which were upregulated in TAMs versus developmental macrophages, and in late stage TAMs versus TAMs from the earlier premalignant stage of breast cancer. From this list of genes upregulated in TAMs, candidate cell surface macrophage markers that could be used to classify TAMs based on there association with one of the distinct tumor microenvironments was selected. These candidate markers will be used to visualize macrophages associated with different regions within the tumor microenvironment using immuno-staining of tumor sections. In preliminary work, CD16 was selected as one potential candidate gene that was upregulated late in tumor development compared to earlier stages. Immunostaining indicated CD16 was expressed on a set of macrophages near blood vessels near the tumors, but not in macrophages located within tumors. Further evaluation of these markers would facilitate isolation of different subsets of macrophages within tumor and understanding the function of these macrophages in tumor microenvironment. Another major cell group involved in all the changes in the tumor microenvironment discussed is the fibroblast. Originally the fibroblast inhibits the growth of the tumor. However through natural selection tumors evolve to change the microenvironment and like wise the function the fibroblast are performing to aid in tumor progression rather than suppress tumor progression. These fibroblast reprogrammed by the tumor cells are called tumor associated fibroblast TAFs. TAFs aid in tumor progression by angiogenesis, immune suppression, and matrix remodeling, providing growth and anti apoptotic factors and aid in metastasis by recruiting TAMs. A key gene involved with this is IKK. It is our hypothesis that ablating IKK in the fibroblast will lead to a reduction of tumor growth.

This document presents the results of the monitoring of a repaired coral reef injured by the M/V Connected vessel grounding incident of March 27, 2001. This grounding occurred in Florida state waters within the boundaries of the Florida Keys National Marine Sanctuary (FKNMS).The monitoring program at the Connected site was to have included an assessment of the structural stability of installed restoration modules and biological condition of reattached corals performed on the following schedule: immediately (i.e., baseline), 1, 3, and 6 years after restoration and following a catastrophic event. Restoration of this site was completed on July 20, 2001. Due to unavoidable delays in the settlement of the case, the “baseline” monitoring event for this site occurred in July 2004. The catastrophic monitoring event occurred on August 31, 2004, some 2 ½ weeks after the passage of Hurricane Charley which passed nearby, almost directly over the Dry Tortugas. In September 2005, the year one monitoring event occurred shortly after the passage of Hurricane Katrina, some 70 km to the NW. This report presents the results of all three monitoring events
