Certain pathogenic Escherichia coli known as Shiga toxin (Stx)-producing Escherichia coli (STEC) are commensals in cattle, and typically cause bloody diarrhea in humans once the Stx toxin is secreted in invaded intestinal epithelial cells. Infections with STEC cells can lead to hemolytic uremic syndrome, which is commonly associated with kidney failure. Several STEC serogroups have been declared adulterants in raw, non-intact ground meat, and future regulations could potentially lead to a higher number of STEC serogroup detection strategies for these pathogenic microorganisms. Microbiological research laboratories may benefit from formalin-fixed STEC cells for periodic (daily, weekly, monthly, among others) instrument validation/calibration by serving as a working set of known cell concentration samples and internal standard i.e. positive control. These cell concentrations may be used across laboratories in different geographical locations, within an individual laboratory, and across a broad range of detection assays (molecular as well as immuno-based). This thesis consists of three research parts: a comprehensive literature review that covers STEC incidence in foods and molecular detection techniques (chapter 1), a literature review that covers immuno-based detection strategies (chapter 2), and a research manuscript that involves the development of an internal standard and positive control with formalin-fixed STEC cells that can be used for a broad range of molecular as well immuno-based detection assays for instrument calibration and validation purposes (chapter 3)

In-situ Analysis of Polymer Film Coats Using AFM and Thermal Probe Methods
The in-situ analysis of pharmaceutical solid dosage forms films is of great interest to pharmaceutical research, especially with the drive towards continuous processing compared to batch processing requiring either in-line or on-line analysis of pharmaceuticals. The size and shape of many solid dosage forms impedes their in-situ analysis, particularly when using thermal analysis techniques such as differential scanning calorimetry (DSC). Thermal probe methods, which combine atomic force microscopy (AFM) with thermal analysis, permits imaging and characterisation of solid dosage forms in-situ. The goal of this thesis is to perform the in-situ analysis of film coated minitablets through the use of thermal probe methods. Minitablets, which are small diameter tablets, were coated with Opadry I, Opadry II or Surelease, and blends of Surelease and Opadry I and Surelease and Opadry II. In addition the same systems were prepared as cast free films to ascertain differences resulting from the two methods of producing the films. Distinct morphologies were observed between the film coated minitablets and cast free films, particularly for the polymer blends. Thermal analysis by modulated temperature DSC (MTDSC) and localised thermomechanical analysis (L-TMA) of film coated minitablets coated with polymer blends indicating miscibility. However the novel technique of heated tip tapping mode was employed, which was able to resolve separate domains of the polymer blends, which appeared as nanophases. Heated tip tapping mode was further employed to determine changes in the surface morphology of film coated minitablets which had undergone curing. It was successfully used to observe the further coalescence of Surelease and the alteration in the phase distribution of the polymer blends after film coated minitablets had been cured. Polymer films with incorporated pigments were also analysed by thermal probe methods with differences in pigment distribution being observed between film coated minitablets and cast free films. Tapping mode AFM was able to determine the location of pigment particles and the novel use of L-TMA performed in tapping mode allowed thermal analysis without changing AFM mode. Overall the use of AFM combined with thermal probes achieved the aim of performing in-situ analysis of polymer film coats; however further research to correlate in-vivo behaviour of film coats would be of value.

"Massification, a phenomenon driven by both the collective aspirations of citizens and governmental encouragement of a highly skilled workforce, is revolutionising the field of higher education. The student body, particularly in widened participation universities, has expanded in number and diversity increasing the demands on HE institutions and their teaching staff. Australian business school graduates are expected to drive national success in fast-changing, globally competitive business environments. This qualitative study investigates characteristics of teaching excellence in widened-participation teaching environments in Australian business schools. The recency of the massification phenomenon has meant theoretical conceptualisations are sparse so Grounded Theory was used to generate new theory. Using university entry scores as a surrogate for an academically inclusive student cohort, 23 award winning teachers from the business schools of eight universities were interviewed. These exemplary business educators displayed strong similarities in terms of their approach to teaching. Seven dimensions of diversity that significantly impact teaching within a widened-participation environment were identified. This thesis argues that alignment between the diversity of cohort and both the type of educators and the pedagogic strategies they employ is the key to unlocking teaching excellence within widened-participation universities, thus giving rise to the notion of context mediated pedagogy and the associated conceptual model. Findings from this study are significant because extreme student diversity is now the norm in many university environments and a better understanding of teaching excellence may offer specific insights for policymakers, academic leaders and educators seeking to adapt pedagogy for the massified HE environment. Improving teaching quality in these widened-participation universities has the power to significantly impact on individual student success, whilst driving innovation on the global stage." -- Abstract.
