Dans le système nerveux central, les récepteurs couplés aux protéines G (RCPG), après synthèse dans le cytoplasme sont principalement adressés à la membrane plasmique des neurones où ils interagissent avec les neurotransmetteurs. De nombreuses données in vitro suggèrent que l’abondance membranaire en RCPG conditionne la réactivité neuronale à la stimulation. Les RCPG sont, pour la plupart, rapidement internalisés après stimulation par leur ligand. L’étude de ce phénomène s’est particulièrement développée depuis une quinzaine d’années et a engendré une masse considérable d’informations. Pourtant la dynamique de la plupart des RCPG dans les neurones reste mal connue. Dans ce contexte, nos travaux de thèse ont porté sur la dynamique du trafic intraneuronal du récepteur muscarinique m2 (Rm2) dans des neurones d’hippocampe in vitro. Nous nous sommes posé deux questions principales : 1) Quelle est la voie d’endocytose précoce utilisée par le Rm2 après stimulation par un agoniste des récepteurs muscariniques ? et 2) Quel est le devenir post-endocytotique du Rm2 ? Pour répondre à ces questions, nous avons développé des approches méthodologiques adaptées. Nous avons réalisé et validé des constructions exprimant, par transfection dans des neurones d’hippocampe, le Rm2 sauvage ainsi que le Rm2 couplé à la GFP ou la super ecliptic Phluorin (SEP), une GFP pH-dépendante. Nous avons mis au point une approche d’étude dynamique du Rm2 sur neurones vivants utilisant la microscopie confocale “spinning disk”. Nous avons montré que très rapidement après le début de la stimulation par un agoniste muscarinique, le Rm2 s’internalisait dans le neurone en empruntant la voie des puits couverts de clathrine. De plus, nos études sur neurones vivants nous ont permis de déterminer que le Rm2 est endocyté dans des puits pré-existants et non dans des puits néo-formés. Nos résultats montrent pour la première fois que le Rm2 s’endocytose dans le neurone par la voie clathrine dépendante.
