Genetic Variant Rs2383207 is Associated with Susceptibility to Large Artery Atherosclerosis Ischemic Stroke in Brazilian Population
BACKGROUND: Stroke constitutes a severe and complex disorder with high morbidity and mortality rates worldwide. In recent years, genetic factors have been pointed as important risk contributors for stroke pathogenesis. CDKN2A/2B gene has been described in association with the risk of multiple diseases, including cardiovascular diseases, hypertension, and stroke. We aimed to investigate the association of rs2383207 polymorphism, nearby CDKN2A/2B gene, with the susceptibility to Large Artery Atherosclerosis Ischemic stroke (LAA-IS) in Brazilian population. METHODS: For the study, 195 patients classified by TOAST criteria, 249 stroke-free controls from Joinville Stroke Biobank were matched by age and gender. Genotypes were determined by TaqMan real-time PCR assay. RESULTS: We found a significant difference in the distribution of the rs2383207*A allele frequencies between cases and controls (58% vs 19%; p = 4.66 E-10) and increased risk for LAA-IS due to this allele [OR 2.35 (95% CI = 1.79 - 3.08)]. CONCLUSION: To our knowledge, this is the first study correlating the polymorphism rs2383207 with stroke in Brazilian population, which may be useful for understand the genetic role in LAA-IS predisposition.Key- Words: Ischemic Stroke - genetic variants - CKDN2A/2B gene.

I. F. ecuadorensis - taxonomic treatment. Especially diverse in the Andes, F. ecuadorensis is one of the largest genera of grammitid ferns in the Neotropics. It is represented by 49 species, which grow primarily as epiphytes in tropical forests at high elevations. / II. Melpomene - systematic revision. Twenty-nine neotropical species and 10 varieties are recognized. Occurring at 400-5200m elevation in varied habitats, distribution is Mesoamerica, the Caribbean, and tropical South America, with one disjunction to Africa, Madagascar, and the Mascarenes. / III. Alansmia - taxonomic monograph. Twenty-five species and 2 varieties of this primarily neotropical, epiphytic genus are presented. Occurrence is predominantly in montane forests but also lowland forests and páramos.

Cost-utility Analysis of Imaging for Surveillance and Diagnosis of Hepatocellular Carcinoma
Hepatocellular carcinoma (HCC) is a serious complication associated with chronic liver disease. Current guidelines recommended HCC surveillance using ultrasound (US) every six months. However, US surveillance can be challenging for some patients, particularly those with cirrhosis or obesity. Alternately, computed tomography (CT), magnetic resonance imaging (MRI), and abbreviated MRI have been explored as alternative imaging modalities and may be used in selected patients who are likely to have experienced inadequate US examinations. In this thesis, we aimed to assess the cost-effectiveness of imaging-based surveillance and diagnostic strategies in patients at risk of HCC while taking into account technically inadequate examinations and patients'' compliance. We compared seven surveillance and diagnostic strategies: strategy A, US for surveillance and CT for diagnosis; strategy B, US for surveillance and MRI for diagnosis; strategy C, US for surveillance and CT for inadequate or positive surveillance; strategy D, US for surveillance and MRI for inadequate or positive surveillance; strategy E, surveillance and diagnosis with CT followed by MRI for inadequate surveillance; strategy F, surveillance and diagnosis with MRI followed by CT for inadequate surveillance; and strategy G, surveillance with abbreviated MRI followed by CT for inadequate surveillance or MRI for positive surveillance. Two compliance scenarios were evaluated: optimal and conservative. For each scenario, the most cost-effective strategy was based on a willingness-to-pay threshold of Can$50,000 (Canadian dollars) per quality-adjusted life year (QALY). We also performed sensitivity analyses. Our results demonstrated that strategy E was the most cost-effective strategy in scenarios with optimal patient compliance (Can$13,631/QALY). However, in scenarios with low patient compliance, strategy G was the most cost-effective alternative to the current US-surveillance practice (Can$39,681/QALY). Sensitivity analyses supported the base-case analysis in the optimal compliance scenario; however, several parameters altered the cost-effectiveness relationship in the conservative compliance scenario. By performing this economic analysis, we concluded that an individual approach, considering the clinical particularities of the patients, is more cost-effective than the current "one-size-fits-all" strategy. However, before implementing a surveillance program incorporating imaging modalities other than US, future studies should address the economic burden associated with false-positive HCC diagnoses and the accuracy of abbreviated MRI examinations in a surveillance setting.

A Seed Resource for Screening Functionally Redundant Genes & Isolation of Novel Mutants Impaired in Abiotic Responses
Overview: The global atmospheric CO2 concentration is rising and in 2013 it surpassed 400 parts per million for the first time in human history. Plants can sense CO2 concentrations in leaves and adapt transpiration accordingly. Ultimately, plants control gas exchange by opening and closing stomatal pores, which are highly responsive to changes in CO2 concentration, and CO2-mediated regulation of stomatal conductance has profound effects on plants. Among several signals that regulate stomatal movements, the stress hormone Abscisic Acid (ABA) plays a critical role in plant responses to abiotic stresses such as drought. The molecular signaling mechanisms and network principles by which CO2 and ABA control plant gas exchange were investigated. Three unbiased forward genetic screens were pursued and, after screening over 10,000 artificial microRNA lines and over 1,000 EMS mutagenized Brachypodium lines, partially ABA or CO2 insensitive lines were isolated and are currently being characterized. Major Goal: Due to the rising atmospheric CO2 concentration, CO2 control of gas exchange in plants will have profound effects on global water-transpiration, plant water use efficiency, leaf heat stress and optimal stomatal conductance of plants, including crops. Elevated CO2 concentrations in leaves cause stomatal closure, whereas reduced CO2 concentrations result in stomatal opening. However, little is known about the molecular signal transduction mechanisms that mediate CO2-induced stomatal movements, especially in grasses. Besides, the stress hormone ABA plays a key role in drought-induced stomatal closure, and the molecular pathways of CO2 and ABA converge, fine tuning stomatal movements in plants. My main goal was to identify and characterize new genes/proteins that play a role in CO2 and ABA signaling in plants, further elucidating these complex molecular pathways. Specific Goals: Three extensive forward genetic screens were pursued: 1- over 10,000 amiRNA lines were screened for ABA sensitivity, leading to the isolation of partially insensitive lines, 2- over 10,000 amiRNA lines were screened for low [CO2] sensitivity, leading to the isolation and characterization of lines and 3- over 1,000 EMS mutagenized Brachypodium lines were screened, leading to the isolation of two [CO2]-insensitive lines. As some of these finding have already been published (Hauser and Ceciliato et al., 2018 Journal of Experimental Botany), the other most relevant lines are being further investigated.
